Abstract:
PROBLEM TO BE SOLVED: To provide a highly bioadherent cultured periodontal ligament cell sheet having satisfactory adherence to a dental root surface.SOLUTION: A cultured periodontal ligament cell sheet that is manufactured by: culturing a cell under a specific condition on a cell culture support in which a substrate surface is coated by temperature-responsive polymer whose upper limit or lower limit critical dissolution temperature to water is 0 to 80°C; (1) setting culture solution temperature to the upper limit critical dissolution temperature or higher, or the lower limit critical dissolution temperature or lower; (2) closely adhering a cultured periodontal ligament cell sheet to a carrier; and (3) peeling the sheet together with the carrier, has a ligament-like microstructure, extremely high bioadherence to a dental root surface, can transplant a target cell highly densely, and can positively reconstruct periodontal tissue. Furthermore, by laminating a cell sheet to be transplanted to allow the sheet to have three-dimensional polarity, an attachment organ can be reconstructed even more efficiently, and clinical application to middle-level periodontitis, serious periodontitis, gingival recession or the like is strongly expected.SELECTED DRAWING: None
