Abstract:
PURPOSE: To provide a new cyclohexanetriol derivative useful as an intermediate for 1α-hydroxyvitamin D derivatives.
CONSTITUTION: The compound of the formula [R1-R3 are each H or OH- protecting group; X is O, CHCH2OR4, CHCHO, or CHCO2R5 (R4 is H or OH- protecting group; R5 is lower alkyl) ; Y is H; Z is OR6 (R6 is the same as R4); or, Y and Z are combined into single bond, X and Z into =N)-, =CHCH(OR7))-, or =CHCO2- (R7 is H or lower alkyl)], e.g. 1,5,6,7,8,8a- hexahydro-6-methoxymethoxy-3-methoxy-3H-2-benzopyran-7,8-diol. The compound of the formula is useful as an A-ring constituent in synthesizing 1α- hydroxyvitamin D derivative bearing a substituent, esp. at 2-site, for example, being useful for synthesizing 2β-hydroxypropoxy-1α, 25-dihydroxyvitamin D3 promising its clinical applications as a medicine for osteoporosis high in blood sustainability.
