Abstract:

To provide very simple detection of expression of an extraneous gene in a living body while making the extraneous gene live even after the detection, by observing gene expression in the living body after the extraneous gene outside a cell is introduced into a cell of the living body by impressing momentarily the cell with a voltage.

This very simple detection of expression of an extraneous gene in a living body comprises drilling a 1-mm diameter hole in the obtuser edge of an egg shell of a fertilized chicken egg after disinfecting the edge, removing a membrane under the shell, the chorion covering the chicken embryo and the albumen, promptly injecting to the exposed chicken embryo a expression- vector solution that contains a structural gene combined with a base sequence coding a secretary signal peptide of bovine α-lactoalbumin on the upstream side of a firefly luciferase gene, and an enhancer promotor by using a fine glass tube having a polished tip, momentarily impressing the embryo with a voltage of 25V to introduce the extraneous gene into the living body cell while manipulating two electrodes of an electropolator so as to hold the chicken embryo between the electrode, and observing the gene expression in the living body cell.
