Abstract:

PURPOSE: To obtain a DNA a mature human growth hormone, in relation to a method for producing a protein in a bacterium, in detail, operabily ligated to a DNA encoding a signal sequence of alkaliphosphatase or enterotoxin.
CONSTITUTION: A eukaryotic protein is expressed by the following method to secrete a eukaryotic periplasmic space. (a) A vector for expressing a direct hybrid to be secreted, containing a sequence in which 3' terminal of DNA encoding a eukaryotic secretion signal sequence and is bonded to 5' terminal of a DNA encoding a mature protein, is constructed, (b) a eukaryotic host molding organism is transformed with the vector of the process (a), (c) the host transformed by the process (b) is cultured and (d) a mature protein is accumulated in periplasm of the host.
