A nucleotide monophosphate (e.g. deoxythymidine monophosphate) as a substrate, phosphoenol pyruvic acid, adenosite triphosphate(ATP), sucrose, and pyruvic acid kinase, sucrose synthase and deoxythymidine-D- glucose 4,6-dehydratease as enzymes are simultaneously brought into contact with each other and incubated in a buffer solution by a batch method or by a continuous method to obtain the objective nucleotide-6-deoxy-D-xylo-4-hexulose useful for producing tetrasaccharide sialyl-Lewis X or its derivative, etc., at a low cost by the enzymatic synthesis method.
