Abstract:

PURPOSE: To obtain an optically active substance useful as pharmaceuticals, etc., in high efficiency, by carrying out ester interchange reaction of a specific ester with an alcohol in an organic solvent containing suspending enzymes.

CONSTITUTION: A suspension having a water-content of ≤2W/V% is produced by mixing (A) an ester of a straight-chain or branched-chain aliphatic racemic alcohol having a principal structure of formula I (R1W7 are functional groups containing one or more elements such as H, C, halogen, O, N, S, P, etc.; C2 is asymmetric carbon; two or more of R1W3 are same functional groups; R6 and R7 are same functional groups) (e.g. 2,2-dimethyl-1,3-dioxolane-2-methanol ester), (B) a 3W6C straight-chain aliphatic alcohol free from asymmetric carbon atom and (C) a non-aqueous organic solvent such as alcohol, etc. Dried lipase, etc., is added to the suspension and subjected to the reaction of formula II (* represents optically active substance; RS is racemic compound; *R is antipode having absolute configuration of R; *S is antipode having absolute configuration of S) and an optically active substance is separated from the produced reaction mixture.
