PURPOSE: To obtain a new mammal cell useful in producing both subunits of heteropolymeric protein in the same cell, receiving post-translational modification in a single culture and producing a biologically active heteropolymer protein.
CONSTITUTION: This mammal cell is transfected with one or more expression vectors (including a heterogeneous DNA encoding each subunit) composed of a plurality of subunits and capable of producing a biologically active protein hormone modified by post-translational glycosylation, etc. Plasmid pRF375 encoding αhcG and βhcG under the control of mouse metallothioneine promoter is mixed with BPV plasmid of pRF398 and mouse C127 is subjected to transfection with the plasmid mixture by using salmon sperm DNA as a carrier to produce biologically active and translational modified heterodimer hcG. Preparation of each plasmid and transfection to mammal cell are carried out by a recombinant engineering technique.
