PURPOSE: To fix steadily with high reproducibility ENA and Sm antigens by fixing on a solid-phase carrier through amino radical or thiol radical containing substance and bivalent reagent.
CONSTITUTION: Solid-phase carriers, such as plastic material EIA microplate, polystylene-hall, glass beads, etc. and substances containing amino or thiol radical, such as poly-L-lysine, etc. are brought into contact to be combined in a solution, such as phosphoric acid buffer physiological salt solution. Adding ENA antigen or Sm antigen to the solution, and adding a bivalent reagent, a substance containing amino or thiol radical combined with a solid-phase carrier and ENA or Sm autigen is combined by covalent linkage through a biaolent reagent. By bringing the fixed ENA antigen into contact with a human-serum, through an enzym-immuno-assay actuating anti-human immuno-globulin which is identified by enzym the anti-human and anti-Sm antigen can be measured quantitatively.
