Abstract:

PROBLEM TO BE SOLVED: To provide a screening method for a substance interfering with the association of DOCK2 and ELMO1, a screening method for a substance interfering with the association of ELMO1 and Tiam1 or a method for searching a treatment medicine for an immune related disease such as allergy, autommunity, GvH, graft rejection or the like.
SOLUTION: It is found out that an Rac activation capacity is markedly lowered in a DOCK2 mutant lacking of the 504 amino acid residue of the N terminal of DOCK2 and actin polymerization can not be brought about, to identify MLMO1 as a molecule bonded to this region. It is found out that DOCK2 is associated with ELMO1 through an SH3 domain. Further, it is found out that ELMO1 is bonded to Tiam1 functioning as an Rac specific GDP/GTP exchange factor (GEF) and it is found out that Rac is activated by recruiting Tiam1 by DOCK2 through ELMO1.
