A 41kDa N terminus fragment constitutes the recognition domain of the FokI restriction endonuclease and the 41kDa C terminus fragment constitutes a nuclease domain of FokI restriction endonuclease. A hybrid restriction enzyme contains a nuclease domain of the FokI restriction endonuclease connected with a recognition domain of another enzyme. Ubx-FN is one of such hybrid restriction enzymes. The enzyme contains a Ubx homeodomain connected with the FokI cleavage or nuclease domain. The invention also relates to two insertion mutants of a FokI endonuclease.
