Abstract:

PURPOSE: To efficiently produce glucosyltransferase by transforming a microorganism to be integrated with a DNA containing glucosyltransferase gene and culturing the transformant.

CONSTITUTION: A recombinant DNA integrated with a DNA containing glucosyltransferase gene, such as pSa123 which is a transformed vector. A microorganism is transformed with the transformed vector, the transformed microorganism is cultured by an ordinary culture method to give a culture mixture, which is filtered or centrifuged to collect a culture solution. Then glucosyltransferase is purified from the culture solution by using salting out with ammonium sulfate, centrifugal separation, desalting and various kinds of chromatographies and collected.
