PURPOSE: To obtain the subject new compound being a specific heterocyclic carbonyl compound, having high inhibiting action having high selectivity to thrombin and useful as a diagnostic for measuring thrombin in blood, an antithrombotic agent, a preventing and treating agent for thrombosis.
CONSTITUTION: This new heterocyclic carbonyl compound is expressed by formula I [A is a 5- or 6-membered monocyclic hetrocylcle having one or more N, a bicyclic heterocycle to which a benzene ring or pyridine ring is condensed; R1 is a 1-4C alkyl, guanidino, amino, OH, SH, a (substituted)phenyl, etc.; B is a group, etc., of formula II (R3 is H, an alkyl, phenyl, etc.); R2 is methyl, aryl, etc.; X is H, OH, amino, a halogen, etc.; m is 1-5; n is 0-4] and has thrombin-inhibiting action. The compound is obtained by reacting a carbinol compound of formula IV prepared by reacting an aminoaldehyde compound of formula III (Alloc is an allyloxycarbonyl) with a silyl compound of the formula (CH3)3SiA with a carboxylic acid compound of formula V.
