Abstract:

PROBLEM TO BE SOLVED: To measure the concentration of pg/ml level or below by conjugating a low molecular compound with biotin and binding a fragment of avidin bonded DNA (dioxyribonucleic acid) noncompetitively and then measuring the quantity of DNA thereby determining the low molecular compound.
SOLUTION: A biotin derivative is conjugated directly or through a crosslinking reagent with a low molecular compound and then the low molecular compound is bonded with an anti-low molecular compound antibody, i.e., a bonded carrier of antibody obtained by bonding a low molecular compound bonded or not bonded with a high molecular compound such as bovine serum albumin and then immunizing an animal such as a rabbit. It is then cleaned and an avidin bonded DNA, e.g. a plasmid DNA fragment, is bonded with the biotin part of biotin.low molecularcompound.anti-low molecular compound antibody bonded carrier. Amplification reaction of DNA is caused and the quantity of low molecular compound is determined based on the quantity of DNA thus measuring the low molecular compound with high sensitivity.
