Abstract:

PURPOSE: To produce human superoxide dimustase which is used as an anti- inflammatory, by forming a gene having the DNA sequence of ompA signal peptide on the upper stream of the DNA sequence coding the amino acid sequence of a polypeptide.

CONSTITUTION: A gram negative bacterium containing a gene where a gene coding the DNA for ompA signal peptide is bonded to the upper stream of the DNA sequence coding the amino acid sequence of a protein or polypeptide as a manifestation gene in nonsecretion type cells, is cultured. Thus, the protein or polypeptide is manifested and extracellularly secreted and human superoxide dimustase (h-SOD)is collected. h-SOD is isolated and purified by separating the cell bodies by centrifugation, when the culture mixture reaches high concentration, followed by extraction, ion-exchange chromatography or the like.
