This new recombinant polynucleotide contains at least 15 bases shown by bases 1-2440 of a base sequence of the formula and a human telomerase reverse transcriptase (hTRT) promoter sequence. This nucleic acid encodes a catalytic protein subunit of human telomerase and a polypeptide related to it. The polynucleotide and the polypeptide are effective as a compound for diagnosing a human disease, its prognosis, treatment, changing proliferation ability of cell and organism and treating diseases such as a cancer, identifying treatment, screening, etc. The nucleic acid is obtained by screening a human fibroblast genome library to identify a genome containing a hTRT RNA code sequence and binding the genome to a λ phage base or the like.
