PURPOSE: To enable the specifying of a coupling form of a DNA reactant on DNA by a method wherein a ruthenium complex is dispersed into a sample liquid, a current is applied to the liquid to cause an electrochemical luminescence and then, the state of the luminescence is detected.
CONSTITUTION: A ruthenium complex causes luminescence electrochemically in a sample liquid with the coexistence of DNA. Here, the level of the luminescence increases with an increase in the amount of a DNA reactant (chemicals such as carcinostatic), it is determined that the DNA reactant is coupled to a major group of DNA. When the level does not increase in substance, it is determined that the DNA reactant is coupled to a substance other than the major group. It is preferable that the ruthenium complex herein used has a phenanthroline based compound such as Ru(phen)32+ as ligand. The coexistence of oxalate in the sample liquid enables increase in the level of the luminescence of the ruthenium complex. As the method to apply the current to the sample liquid, the use of a potential step method allows the obtaining of the luminescence stable and sufficient.
