To provide a method for identifying an agonist and an antagonist, concerning tumor necrosis-related receptors TR1, TR3 and TR5, by contacting a candidate compound with each of TR1, TR3, and TR5, in the presence of ligand TL3. followed by evaluating a binding ability with each of TRs under competition with TL3.
This method comprises the steps of (1) contacting a candidate compound with each of TR1, TR3, and TR5, in the presence of ligand TL3 to a labelled or unlabelled tumor necrosis factor receptor (TNF-R)-related polypeptide, and then (2) evaluating a binding ability of the candidate compound with each of TR1, TR3, and TR5, in competition with TL3. An agonist or antagonist against TR1, TR3 or TR5, is identified by analyzing whether the candidate compound affects a signal generated by TR1, TR3, or TR5 polypeptide located on cell surface or not, followed by determining in the following criteria: a candidate compound promoting generation of the signal is an agonist; a candidate compound inhibting generation of the signal an antagonist.
