PURPOSE: To perform the quantitative assay of a substance to be examined with high accuracy, by preliminarily mixing a specific pretreatment reagent with a specimen to be examined and a complement to perform reaction prior to reacting a lyposome reagent and removing the non-specific lytic factors contained in said specimen and complement.
CONSTITUTION: Prior to performing quantitative assaying by a lyposome reagent, the treatment by a pretreatment reagent is preliminarily applied to a specimen and a complement. This pretreatment reagent has the same constitution as the actual lyposome constituting the lyposome reagent used in the quantitative assaying of a substance to be examined, that is, consisting of at least one of phospholipid and glycolipid and cholesterol and further comprises a lyposome containing a crosslinking agent but has no marker substance sealed therein. Each of phospholipid, glycolipid, cholesterol and the crosslinking agent is pref. has the same diameter as the lyposome reagent and said components are pref. contained in the same ratio. By applying the treatment by thus constituted pretreatment reagent, the effect of a non-specific dissolving factor destructing the lyposome reagent other than the substance to be examined and flowing out the sealed marker substance can be removed. Therefore, the substance to be examined can be subjected to quantitative assaying with high accuracy.
