Abstract:

PURPOSE: To obtain new indane derivs. and quinoline derivs. which have an effect to activate a potassium channel and are useful as a drug for cardiovascular diseases and as an antiischemic agent.

CONSTITUTION: The compds. include a compd. expressed by formula I or its pharmacologically acceptable salts. In formula I, X is O, S, NCN, and when X is O or S, A is a single bond, while, when X is NCN, A is a single bond, CH2, NR9 (R9 is H, a 1 to 4C alkyl), S, SO, SO2; Y is NR8 (R8 is H, an alkyl, etc.), O, S, etc.; R1 is an aryl, an arylalkyl, etc.; R2 is H, OH, etc.; R3 and R4 are H, an alkyl, etc.; R5 is H, an alkyl, a haloalkyl; R6 is H, an alkyl, a halo, OH, etc.; R7 is R8, for example, (trans)-N"-cyano-N-(2-hydroxy-3,3-dimethyl-6- nitro-1 indanyl)-N'-phenylglyanidine. For example, the compd. expressed by formula I in which X is O and Y is NR8 is obtd. by allowing a compd. expressed by formula II to react with 4-nitrophenylchloroformate to obtain a compd. expressed by formula III and then allowing the obtd. compd. to react with an amine expressed by formula IV.
