Abstract:

PROBLEM TO BE SOLVED: To provide an integrated method and device for the amplification and detection of nucleic acid. SOLUTION: Nucleic acid is detected by (a) preparing a sample for the detection of nucleic acid, (b) amplifying the nucleic acid in a device in the presence of at least one kind of nucleic acid primer having specificity to the nucleic acid to be detected and at least one kind of capturing probe having specificity to the nucleic acid to be detected wherein said capturing probe is fixed to at least one inner surface of said device, the capturing probe is selected to form a hybrid of the nucleic acid and the probe having a melting point lower than the melting point of the hybrid of the nucleic acid and the primer and the amplification is carried out under a condition under which the hybrid of the nucleic acid and the primer is present in stable state and that of the nucleic acid and the capturing probe is unstable and (c) bonding the amplification product to the fixed capturing probe.
