Abstract:
PURPOSE: To obtain a new KO-8119 substance effective against drug-resistant Coccidium and useful as a therapeutic agent, etc., for treating coccidiomycosis, by culturing a microorganism which belongs to the genus Streptomyces and is capable of producing KO-8189 substance in a medium.
CONSTITUTION: A microorganism [e.g. Streptomyces sp. KO-8119 (FERM P-13398)] which belongs to the genus Streptomyces and is capable of producing KO-8119 substance expressed by formula I (R is a group of formula II, formula III, etc.) is inoculated into a medium and cultured in a jar incubator at 27°C for 3 day to afford a seed culture medium. Then, the seed culture medium is subjected to submerged culture at 27°C for 3 day and pH of the cultured mixture is adjusted to 8.5 and the cultured mixture is extracted with ethyl acetate and the extracted liquid is concentrated and purified by silica gel chromatography and reverse phase high-performance liquid chromatography to provide the objective new KO-8119 substance expressed by formula I, effective against drug-resistant coccidium, especially coccidium having resistance to polyether-based compound.
