To provide a liquid cultivation method of Schizophyllum commune Fr. to separate β-1,6-branched-β-1,3-glucan, and further to prepare a preparation composition for external use, containing the β-1,6-branched-β-1,3-glucan produced by the method, and usable for prevention of skin aging, treatment of skin damage, and whitening of the skin.

This liquid cultivation method of Schizophyllum commune Fr. to separate β-1,6-branched-β-1,3-glucan comprises a step for preparing a mycelium of the Schizophyllum commune Fr., a step for culturing the prepared mycelium in a liquid medium containing 1-10% glucose, 0.1-1% yeast extract, 0.05-0.5% ammonium diphosphate or ammonium sulfate, 0.05-0.5% potassium monophosphate and 0.005-0.2% magnesium sulfate, and regulated so as to have pH 4.0-8.5, a step for separating the produced β-1,6-branched-β-1,3-glucan from the liquid medium, and a step for purifying the obtained β-1,6-branched-β-1,3- glucan.
