PURPOSE: To provide a local ophthalmic composition useful in the treatment of glaucoma and ocular hypertension which shows excellent reduction in the intraocular pressure without causing significant side-effects.
CONSTITUTION: A local ophthalmic composition for the treatment of glaucoma and hypertension comprises a therapeutically effective amount of cloprostenol or fluprostenol of the formula [wherein R1 is H, 1-12C alkyl, 1-12C acyl, 3-8C cycloalkyl, a cationic salt or a pharmaceutically acceptable amine; R2 and R3 are each H, 1-5C alkyl or R2 and R3 together are O; X is O, S or CH2; the composite portion of a solid line and a broken line is a single bond, a cis- or trans-double bond with α-chain (upper chain) and a single bond or a trans- double bond with ω-chain (lower chain); R9 and R11 are each H, 1-10C alkyl or 1-10C acyl; Y is O or in each configuration H and OR15 (R15 is H or 1-10C alkyl) or 1-10C acyl; and Z is Cl or CF3] or an analogue thereof. The concentration of said compound in the composition is in the range of 0.0003-0.3 wt.%, preferably 0.003-0.03 wt.%.
