Abstract:

PURPOSE: To obtain a new enzyme useful for converting maltose originated from starch into trehalose and a glucide containing trehalose in high conversion.
CONSTITUTION: This maltose.trehalose conversion enzyme has an action to convert maltose into trehalose and trehalose into maltose. It has a molecular weight of about 57,000-120,000 dalton (SDS-gel electrophoresis) and an isoelectric point pI of about 3.8-5.1 (ampholine-containing electrophoresis) and is inhibited by 1mM Cu2+, Hg2+ or 50 mM tris-HC1 buffer solution. The enzyme produced by a microbial strain of the genus Pimelobacter has the optimum temperature of about 20°C (pH 7.0, 60min) and the optimum pH of about 7.0-8.0 (25°C, 60min) and is stable at about ≤30°C (pH 7.0, 60min) and at pH 6.0-9.5 or thereabout (20°C, 60min). It can be produced by culturing Pimelobacter sp. R48 (FERM BP-4315) in a nutrient medium and collecting from the cultured product.
