To make it possible to measure scarified hemoglobin A1c antibody simply and quickly by providing an organic silicon film on the metal film on a transparent substrate, and using a surface plasmon resonance biosensor having a hemoglobin A1 antibody on the film.
A metal film 2 of gold, silver, copper or the like is formed on a transparent substrate 1, which is transparent for laser light such ad glass polyethylene terephthalate and polycarbonate, by sputtering, evaporation and the like. On the metal film 2, an organic silicon film 3 containing Si-O and SiC coupling in molecules is formed by using a silane coupling agent and the like. On the organic silicon film 3, HbA1c 4 is fixed. The hydrolysis group in the silane coupling agent is coupled with the metal atoms in the metal film 2, and the organic functional group is coupled with the metal atoms in the metal film 2, and the organic functional group is coupled with the HbA1c 4. Therefore, the three members of the metal film 2, the organic silicon film 3 and the HbA1c 4 are rigidly coupled. Thus, the HbA1c 4 can be made to be fixed very close to the metal film 2, and the measuring sensitivity can be improved by a large extent.
