Abstract:
The invention relates to a method of inhibiting a function of a cytosolic protein by a single chain antibody (scFv). More specifically, it relates to a method of inhibiting the function of a cytosolic protein by using a single chain antibody constructed by removing a signal peptide of one of VH and VL of an anti-cytosolic protein antibody, a method of analyzing the function of a cytosolic protein by using the single chain antibody, a method of producing the single chain antibody which has a characteristic of specifically inhibiting the function of a cytosolic protein and a non-human transgenic animal in which the single chain antibody which has a characteristic of specifically inhibiting the function of a cytosolic protein is expressed and the function of a cytosolic protein is inhibited. It was revealed that a scFv gene (SHL) in which VH and VL of an anti-WASP-EVH1 monoclonal antibody gene were utilized and VH having a leader sequence and VL in which a leader sequence was removed were connected with a linker and a gene (SHL-CL) having a CL sequence were expressed in the cytosol and inhibited the function of a cytosolic protein.
