To provide a method for isolating a total cellular RNA (tcRNA) from a biological material, capable of decreasing a genomic DNA (gDNA) in a biological sample, without introducing harmful contaminants, nor significantly requiring a time, and to provide a device for the same.
This method comprises, for example, removing a considerable amount of the gDNA, while maintaining integrity of the RNA in the sample. A preliminary filter column filled with at least one layer formed of glass fiber or borosilicic acid fiber is used therein. Further, a tissue/cell-dissolved preparation is prepared, and then the preparation is introduced into the preliminary filter column. When a homogenate passes through the preliminary filter column, the cellular contaminants containing the gDNA remain in the column and, on the other hand, the tcRNA is partially contained in an effluent therefrom. Furthermore, the preliminary filter column is used, before the sample is subjected to an additional purification process or subsequent processes.
