Abstract:

PURPOSE: To easily obtain an erythropoietin clone capable of manifesting biologically active erythropoietin by screening a human embryonic liver-cDNA library by using a specific erythropoietin clone.

CONSTITUTION: This method for cloning human cDNA capable of manifesting erythropoietin is to digest purified erythropoietin protein with trypsin, prepare a pool of oligonucleotide probes based on amino acid arrangements of the obtained trypsin fragments, screen a human genome-DNA library by using the oligonucleotide probe, select a clone capable of hybridizing with the above probe, determine a base arrangement, inspect if the base arrangement is erythropoietin clone or not, screen a cDNA library prepared from the human embryonic liver by using the identified erythropoietin clone and select the objective erythropoietin clone.
