To provide a method for deleting or expressing a specific gene by a Purkinje cell of camphor neurocyte and a bipolar cell of a retina neurocyte.
This method for creating an animal of which cerebellar Purkinje cell and a retina bipolar cell can be genetically manipulated comprises inserting a foreign gene into a chromosome DNA of L7 gene by a gene targeting method and expressing the foreign gene at least in one of the cerebellar Purkinje cell and the retina bipolar cell. Especially, a targeting vector is prepared by inserting a Cre gene into the upstream of the code part of the translation initiation site of the chromosome DNA of the L7 gene. A recombinant ES cell having a modified L7 genetic locus is prepared by subjecting the targeting vector to a homologous recombination method. A chimera animal is prepared by using the ES cell or a Cre animal which is an offspring of the chimera animal and has a modified L7 genetic locus in a somatic cell in a homogenous or heterogeneous state is created. On the other hand, a fLox animal into which a gene containing at least two LoxP sequences are transferred is created and the chimera animal or the Cre animal is hybridized with the fLox animal.
