PURPOSE: To simply detect a specific base sequence by eliminating a procedure for labeling DNA with a chemical substance.
CONSTITUTION: A solution containing a single chain DNA 70 having a specific base sequence of DNA to be detected is mixed with a positive charge gold colloid 72 to prepare a DNA probe 74. A specimen DNA 76 and the DNA probe 74 are put in a temperature controlling bath 78, heated to 90-95°C to cleave the specimen DNA. Then, the temperature is gradually lowered and incubation is carried out at 37-70°C for several minutes to one hour. In the case where a base sequence corresponding to the single chain DNA of the DNA probe 74 is contained in the specimen DNA 74, the single chain DNA 76a of the specimen DNA 76 is hybridized with the single chain DNA of the DNA probe 74 to form a duplex DNA in the DNA probe.
