To provide a method for allowing efficient discrimination of α2,6-sialo-sugar chain included in a biological sample from α2,3-sialo-sugar chain.
Methyl esterification using a 1-methyl-3-p-tolyltriazene (MTT) reagent is applied to a sialo-sugar chain in a sugar chain sample captured in a carrier, and then a sugar chain is isolated. A methyl-esterified α2,3-sialo-sugar chain in the isolated sugar chain is demethylated under an acid condition; reductive amination reaction is performed by reacting the isolated sugar chain with a compound having an amino group to label the sugar chain by the compound having the amino group. By subjecting thus obtained labeled sugar chain to high-speed liquid chromatography or mass analysis, the α2,6-sialo-sugar chain and the α2,3-sialo-sugar chain can be efficiently discriminated from each other.
