Abstract:

To provide a method for readily and rapidly measuring N-myristoyl transferase (NMT) activities in high sensitivity; to provide a kit for measuring the N-myristoyl transferase activities; and to provide a method for screening a material inhibiting the N-myristoyl transferase activities.

The method for measuring the N-myristoyl transferase activities in a sample to be measured comprises (1) a step for carrying out an enzyme reaction by mixing the sample to be measured, containing the N-myristoyl transferase, with a peptide substrate having the N-terminal containing Gly and the C-terminal labeled with a labeling material, and a myristoyl CoA to form an N-myristoylated labeled peptide, (2) a step for capturing the N-myristoylated labeled peptide formed at the reaction by an antibody immobilized on a solid carrier and recognizing an N-Myr-Gly part of the peptide, and (3) a step for detecting the N-myristoylated labeled peptide captured by the reaction by utilizing the labeling material.
