Abstract:

PURPOSE: To measure thyroid-stimulating hormone (TSH) with high sensitivity by using TSH-β subunit specific monoclonal antibodies respectively having different recognition epitopes as an immobilizing antibody and labeling antibody.

CONSTITUTION: The spleen cells of the mouse immunized by TSH or TSH-β subunit is processed to obtain the β subunit specific monoclonal antibody. The TSH-β subunit specific monoclonal antibodies (A) having the different recognition epitopes are obtd. therefrom. For example, the TSH, the antibody A1 labeled with horse radish peroxidase and polystyrene beads immobilized with the antibody A2 are brought into reaction with each other. The antibodies A1 and A2 have the different recognition epitopes. Absorbancy is then measured with the labeled matter after the reaction and the concn. of the TSH is determined. Since the antibodies A having the different recognition epitopes are used, the measurement of the TSH concn. by the one-step method with the high specificity and high sensitivity is made possible.
