Abstract:
PURPOSE: To provide a new method for producing a superoxide dismutase analog having substantially the same amino acid sequence and activity as those of natural human Cu/Zn superoxide dismutase.
CONSTITUTION: This method for producing superoxide dismutase analogs is to proliferate a culture of bacteria cells in a Zn-containing production medium supplemented with Cu++ in an uninhibiting amount for proliferation so that the final Cu++ concentration in the production medium becomes larger than 2 ppm and recover produced and enzymatically active human Cu/Zn superoxide dismutase analogs. The said cells contain and can express DNA coding for the polypeptide analog of human Cu/Zn superoxide dismutase. The cultured product is proliferated in a suitable condition capable of expressing the DNA and producing the polypeptides in bacterial cells.
