Abstract:

To provide a method for screening a substance promoting or inhibiting production of interferon-α through a signal transfer route of a Toll-like receptor 7 (TLR7) or a Toll-like receptor 9 (TLR9) of an interleukin-1 receptor-associated kinase 1 (IRAK-1) which interacts with an interferon regulatory factor 7 (IRF7).

This method comprises expressing the IRAK-1 and the IRF7 in a cell in which a test substance exists and measuring a degree of association of the the IRAK-1 and the IRF7. Further, the method comprises measuring a degree of phosphorylation of the C-terminal of the IRF7 by the IRAK-1 in the presence of the test substance in vitro. Furthermore, the method comprises administering the test substance and a TLR7 ligand or a TLR9 ligand to a model mouse which is unresponsive to the TLR7 ligand or the TLR9 ligand, or to a cell derived from the mouse, or stimulating the mouse or the cell by the substance and the ligand, and measuring an amount of the interferon-α produced in the mouse or in the cell derived therefrom, or a degree of activation of the IRF7 therein.
