PURPOSE: To quickly and efficiently measure pollution of milk by microorganisms by mixing a liquid milk sample with a chelating agent and then centrifuging the sample and separating a cellular component in a pellet distinguished from other milk components.
CONSTITUTION: In a method separating and concentrating cells from a liquid milk sample, a chelating agent (e.g. ethylene diamine tetraacetate) is added to the milk sample and mixed therewith, the sample is centrifuged for a prescribed time and the supernatant containing non-cell milk component is removed from a cell pellet obtained by centrifugation. Cells are separated from the milk components bound by the chelating agent and then the cell pellet is re-suspended in a buffer and a dissolving agent is added to dissolve the cells and luciferase- luciferin reagent is added to the resultant solution. Relative amount of ATP existing in the dissolved sample is determined by measuring a relative photoemitting quantity, emitted by the sample. Thereby pollution of milk by microorganisms is quickly and efficiently measured.
