To provide a method and a system for analyzing gene expression state in biological cells cultured in a pseudobiological environment while suppressing the amount of a medium liquid to a low level.
The system comprises a flow culturing machine 1, an RNA retrieving means 110 and an analyzing means 120. In this system, the flow culturing machine 1 functions to make a culture liquid 47 of high dissolved oxygen concentration flow to the 1st chamber 41 side of a diaphragm 31 disposed with static-cultured HUVEC(human umbilical venous epithelial cell) and make a culture liquid 47a of low dissolved oxygen concentration flow to the 2nd chamber 43 side of the diaphragm 31 and enables HUVEC to be exposed to either one excitation among a culture liquid flow, oxygen gradient-added culture liquid flow, low-specific gravity-lipoprotein-added culture liquid flow and oxygen gradient- and low-specific gravity-lipoprotein-added culture liquid flow. The means 110 functions to retrieve an elution liquid obtained by removing a medium from HUVEC after flow culture and adding an iso-gene to effect cytolysis. The means 120 functions to analyze the expression state of RNA in the retrieved elution liquid by DNA microarray or real-time PCR (polymerase chain reaction) technique.
