Abstract:
PROBLEM TO BE SOLVED: To provide methods and composition that generate single-stranded breaks (nicks) in a double-stranded DNA and facilitate targeted integration by homologous recombination at the nicked site, without simultaneously generating error-prone NHEJ repair in mammalian/human cells.SOLUTION: A protein complex comprises: a fusion protein comprising a DNA-binding domain and at least one catalytically inactive cleavage domain or cleavage half-domain; and a second fusion protein comprising a zinc finger domain and a catalytically active cleavage half domain. The catalytically inactive cleavage half domain forms a heterodimer with the catalytically active cleavage half-domain of the second fusion protein.SELECTED DRAWING: None
