Abstract:

PROBLEM TO BE SOLVED: To obtain a modified nucleotide comprising a specific modified nucleotide having detectable signal, capable of easily being detected when bonded to or enclosed in a nucleic acid material and useful as a probe, etc., for detection and isolation of DNA or RNA.
SOLUTION: This modified nucleotide is a new modified nucleotide of formulas I (P is a phosphate residue; S is residue of a saccharine or a monosaccharide; B is a residue of a base of purine or pyrimidine with the proviso that when the nucleotide is deoxyribonucleotide, the B binds with 3' and/or 5' position of phosphate residue, and when the nucleotide is ribonucleotide, the B binds with 2', 3' and/or 5' position of the phosphate residue; Sig is a chemical residue convalently bonding to a saccharide of a nucleotide, a monosaccharide residue S or a phosphate residue P, and allowing itself to be a signal, further, allowing itself to be detected or capable of informing its existence) or II. The modified nucleotide is useful as a recognizable probe, etc., used for detecting and isolating a DNA and a DNA sequence.
