Abstract:

To provide an improved means for producing in vitro recombinant BMP (bone morphogenic protein) and other TGF-β-family proteins using prokaryotic host and eukaryotic host.

A latent TGF-β-family member fusion protein component for refolding under appropriate refolding conditions is provided, including an N-terminal truncated TGF-β-family protein. The truncated TGF-β-family protein is a fusion protein comprising a TGF-β-family protein C-terminated 7 cysteine domain comprising finger 1 subdomain, finger 2 subdomain and heel subdomain and a breakable modified leader sequence operably joined to the domain and hindering the biological activity associated with the domain, wherein the c-terminated domain is activated in breaking part or the whole of the leader sequence.
