To provide a method for producing a polypeptide that selectively recognizes at least one base pair in a target DNA sequence.SOLUTION: A method includes synthesizing a polypeptide including a repeat domain, in which the repeat domain includes at least one repeating unit derived from a transcription activator-like (TAL) effector, the repeating unit includes a hypervariable region which determines recognition of a base pair in the target DNA sequence, the repeating unit is involved with the recognition of one base pair in the DNA sequence, and the hypervariable region may be (a) HD for recognizing C/G, (b) NI for recognizing A/T, and (c) NG for recognizing T/A.SELECTED DRAWING: Figure 1
