Abstract:
PROBLEM TO BE SOLVED: To provide a method for producing an N-terminal derivative of protein reacting N-terminal of protein or peptide under control conditions to form N-terminal derivative.SOLUTION: The invention provides a method for producing a purified N-terminal derivative of a protein or peptide comprising the steps of: (i) reacting N-terminal amine residue of protein or peptide with a polysaccharide in an acidic aqueous solution to form an N-terminal derivative; and (ii) purifying the obtained N-terminal derivative in an aqueous solution of a pH higher than that in the step (i). The invention also provides N-terminal derivatives of protein or peptide obtainable by the method. The controlled conditions include use of acidic pH for the derivatization step and a higher pH for purification. The derivatives are useful for improvement of pharmacokinetics and pharmacodynamics of protein and peptide.
