Abstract:

PURPOSE: To obtain a chimera oxidase having enhanced activities by expressing a vector, containing a gene having a monoatomic oxygenating active region of a rat hepatic cytochrome P-450 to a reducing activity feeding active region of a reduced nicotinamide adenine dinucleotide phosphate (NADPH)-reductase in a yeast fungus.
CONSTITUTION: A yeast expressing plasmid, containing a chimera oxidase gene constructed by connecting a region required for the monoatomic oxygenating activities of a rat hepatic cytochrome P-450 gene to a region required to manifest the reducing power feeding ability of a rat cytochrome NADPH-cytochrome P-450 reductase gene and capable of expressing the oxidase is transduced into a yeast fungus [e.g. Saccharomyces cerevisiae AH22 (pAMP19)] to transform the yeast fungus. The resultant transformant is then cultured to afford the objective chimera oxidase having the monoatomic oxygenating activities of the cytochrome P-450 and the reducing power feeding ability possessed by the NADPH-cytochrome P-450 reductase and the enhanced monoatomic oxygenating activities.
