Abstract:

PURPOSE: To provide a novel enzyme useful for forming a C-terminal amide group in a physiologically active peptide (calcitonin or secretin) in vitro by the formation of the amide group at the C-terminal.
CONSTITUTION: An enzyme relating to the formation of an amide group at the C-terminal of a C-terminated glycine adduct acts on a C-terminal α- hydroxyglycine adduct of formula I (A is a residual group excluding an α-amino group or imino group and an α-carboxyl group and originated from a natural α-amino acid; X is H, an amino acid derivative residual group bound to the N atom through a carbonyl group) to produce a terminal amide compound of formula II and a glyoxylic acid, has a mol. wt. (by a gel filtration method) of approximately 40000 dalton, an optimal pH of approximately 5-6, a stable pH of 4-9, and a suitable action temperature of 15-35°C. The enzyme can be prepared by treating an enzymatically active substance-containing material such as bovine serum with a substrate-affinitive chromatography having a C- terminal glycine adduct formula III as a ligand and subsequently with an anion exchange chromatography.
