Abstract:

PURPOSE: To obtain a polypeptide such as chloramphenicol acetyl transferase and the like in high yield in high productivity by transforming a host with a specific expression vector, culturing the transformed host, and separating the polypeptide from the culture mixture.
CONSTITUTION: A DNA encoding a polypeptide of prokaryotic or eukaryotic cells effectively binds to and is kept by an expression-controlling DNA sequence that has DNA sequence of the formula and is formed by linking an Escherichia coli phage T5 promoter to an Escherichia coli lac operator or its functional moiety to prepare an expression vector, namely a plasmid that can be replicated in Gram-positive and/or Gram-negative bacterial cells. The host is transformed with the plasmid vector, the transformed strain is cultured under proper conditions, the polypeptide is isolated from the culture mixture whereby the objective polypeptide is obtained.
