NEW MATERIAL: Compd. of formula I {wherein A is formula II [wherein n, o and p are each 0-2; R6 and R7 are each an alkyl, a cycloalkyl or a (substd.) aryl; X is CH or N; Z and W are each O, CHR8 (wherein R8 is an alkyl) or no existence]; R1 is H or an alkyl; R2 is an alkyl, an alkenyl, a cycloalkyl, an aryl, an arylalkyl or the like; R3 is an alkyl, an arylalkyl or the like; R4 is H or an alkyl; R5 is an alkyl, a cycloalkyl, an aryl, a cycloalkylalkyl or the like; and q is 0-2} and their salts and their optical isomers.
USE: A renin inhibitor. Being useful for curing hypertension, conjestive heart failure and other cardiovascular diseases.
PROCESS: E.g. isostere of formula II is reacted on its N-terminal with an amino acid deriv. of formula IV (wherein T is an active group being normally used in peptide synthesis) in an inert solvent to obtain a compd. of formula I.
