Abstract:
PURPOSE: To obtain a new salt which is formed from physostigmine or its derivative having anticholinestersase activity and a phosphatidic acid(s) and is capable of maintaining a high-level pharmaeody namie activity within the brain over a long period by using a method for absorbing the salt into the body through its skin or epidermis, at the time of treating chronic senile dementia. CONSTITUTION: This pharmaceutical composition contains a new salt formed from physostigmine or its pharmacologically acceptable derivative having anticholinesterase activity and a phosphatidic acid(s), or particular a salt represented by the formula (wherein: PS is a cation derived from physostigmine or its pharmacologically acceptable derivative having anticholinesterase activity; PA is a phosphatidic acid or a mixture of different kinds of phosphatidic acid from each other; and x:y is 2:2 to 1:2), such as physostigminedipalmitoylglycerine phosphatidate. This new compound is formed by allowing physostigmine or its pharmacologically acceptable derivative to react with a phosphatidic acid(s) in a solvent such as halogenated hydrocarbon or ketone. The agent is preferably administered in the from of a skin patch, plaster or bandage.
