Abstract:

PROBLEM TO BE SOLVED: To obtain the new subject enzyme excellent in solution stability in the coexistence (absence) of glucose and useful for measuring biological ingredients producing ATP car glucose as products, etc., by culturing a hexokinase producing fungus belonging to the genus Kluyveromyces.
SOLUTION: This new hexokinase is originated from a high temperature yeast belonging to the genus Kluyveromyces, is stable in the coexistence of glucose in a 37°C solution, has a residual activity of at least 80% at 37°C for 7 days and has substrate specificity exhibiting good reactivity with glucose and fructose. The enzyme is useful for measuring ATP or glucose, for measuring biological ingredients (e.g. creatine kinase) producing the ATP or the glucose, etc. The new enzyme is obtained by culturing a high temperature yeast [Kluyveromyces fragilis (IFO 1777), etc.] in a culture medium, separating the product from the culture solution and then purifying the separated product.
