Abstract:

PURPOSE: To provide a new factor multiplying the colony of megakaryocytes in the presence of interleukin-3, having a specific mol.wt., reverse phase high performance liquid chromatography fraction and amino acid sequence, and useful for the treatments of diseases caused by the depression in the function of platelets, etc.
CONSTITUTION: A tumor collected from the lymphonodus of a pancreatic cancer patient is cultured in a medium in an incubator with opening a carbon dioxide line to establish HPC-Y5 strain. The strain is cultured in a serum-free medium, and the cells are separated. The supernatant is collected, concentrated, and subsequently purified by a reverse phase high performance liquid chromatography, etc., to provide the objective megakaryocytemultiplying factor multiplying the colony of the megakaryocytes in the presence of interleukin-3 in vitro, having the single band of a mol.wt. of approximately 32000 by a polyacrylamide gel electrophoresis, eluting with a fraction having an acetonitrile concentration of 40-45% in a 0.1% trifluoroacetic acid and having an amino acid sequence of the formula in the molecule.
