Abstract:

PURPOSE: To provide a new pyrimidinedione derivative having potassium channel opening activity and applicable for the treatment and prevention of disorders caused by the constriction of vascular smooth muscle and other smooth muscle.

CONSTITUTION: The compound of formula I [R1 and R2 are H, 1-6C alkyl or 1-6C alkenyl; R3 and R4 are 1-4C alkyl or together form 2-5C polymethylene; W is H or OH; A is group of formula II (Y1 and Y2 are H, alkylcarbonyl, NH2, NO2, CN, Cl, CF3, alkylsulfonyl, alkoxysulfinyl, alkoxy, cyanomethyl, etc.), 3-pyridyl or 4-pyridyl; X is 1-6C alkyl or NO2], e.g. 6-[4-[3(4- acetylphenoxy)-2-hydroxy-3-methylbutyl]piperazin-1-yl]-1,3-diisopropyl-2,4(1H,3H) pyrimidinedione. The compound of formula I wherein W is OH can be produced by reacting a uracilpiperazine derivative of formula III with a propane oxide derivative of formula IV.
